Brett Spiel Welt Is a good place to start, though it's tough to understand sometimes.

Dominion Isotropic is a good place to play against other humans.

Board Game Arena has a good selection of games to play as well.

BSW doesn't get enough love here. It is the grand-daddy of all the on-line gaming sites, and while a little confusing at first, is pretty easy to get into. Plus, the community is very helpful.

Isotropic is what the OP is looking for. The interface takes a little getting used to, but actually it's pretty good, and they're always improving it. (For instance, in the buy phase it used to be really easy to hit the "play all your money" button without thinking, and you would play Coppers which prevented you from buying Grand Market. But they implemented a fix to avoid this.)

Also they generally get new expansion cards before they go on sale, so that's fun.

Regarding BSW: you want to dedicate the time needed to figure it out. I know it's a headache, I know it's a huge mess, the interface has issues; but it also has TONS of great games. BSW has Dominion, 7 Wonders, Puerto Rico, Settlers, Pandemic, Trans America, Power Grid, and many others.

More importantly, BSW has some amazing players. Personally, I meet some of the best players I know on BSW.

If you're serious about board gaming as a hobby, learn how to use BSW. It will expose you to new games and improve your game play significantly.

It's not as user-friendly, but we play games together all the time through VASSAL on the #boardgames channel on freenode (IRC). See the sidebar for a link. There are also similar things like ZunTzu and Cyberboard, but we haven't played anything through those yet.
